Applications:

Papain-Separopore® 4B can be used as substitute of free papain in many applications. Papain is a cysteine protease that cleaves peptide bonds of basic amino acids, leucine or glycine. It also hydrolyzes esters and amides. This matrix is used to produce Fab fragment of IgG antibodies. It is also used for cell dissociation since it has been shown to be more effective and less damaging with certain tissues. The advantages of using this Papain-Separopore® 4B matrix is that it virtually eliminates autolysis, eliminates contamination of a sample with the protease and allows control of the digestion by removing the papain or controlling the flow of sample over an immobilized papain column. Papain-Separopore® is also more stable against heat-induced denaturation, resulting in longer maintenance of activity. Papain is also used for the study of protein structure and proteomics.

Note: Separopore® is a cost-effective equivalent to Sepharose® in all of its physical properties and binding characteristics.

  • One unit of Papain will hydrolyze 1 μmole of BAEE / min at pH 6.2 at 25 °C

  • Matrix: Cross-linked 4% Agarose beads , Separopore® 4B
  • Activity: 16-40 BAEE units per milligram of papain before immobilization
  • Loading: 250mg papain/ml of settled gel
  • Particle size (range): 63-150 µm
  • Supplied as: 50% slurry in 50% glycerol, 0.1M Sodium acetate ( pH 4.4) containing 0.05% Sodium azide
